I purchased a new Yamaha upright 6 months ago, everthing sound ok until I noticed a problem with the middle E. When playing repeatedly with right pedal pressing, this key does not reponse after a few trikes. Anyone have any idea what is the problem?

Most likely a problem with the front key bushing which requires "easing". Otherwise it would be due to a sluggish action center.

You would need to have a tech come by and then determine exactly what the cause is, however.

I agree with Mark on this one. Have your tech. take a look on the next service. Shouldn't take more than 5 minutes to fix.......actually not more than 2.
